Pyruvic Acid Production from Sucrose by Escherichia coli Pyruvate Dehydrogenase Variants

Sucrose is an abundant, cheap, and renewable carbohydrate which makes it attractive feedstock for the biotechnological production of chemicals. Escherichia coli W, one few safe E. strains able to metabolize sucrose, was examined pyruvate. The repressor csc regulon deleted in W expressing a variant E1 component pyruvate dehydrogenase complex, these were screened shake flask culture formation from sucrose. accumulated at yields 0.23–0.57 g pyruvate/g conversion also accompanied by accumulation some fructose and/or glucose. Selected 1.25 L controlled batch processes with 40 g/L sucrose obtain time–course monosaccharides. Pyruvate re-assimilation observed several strains, demonstrates difference metabolic capabilities glucose- sucrose-grown cultures. An engineered strain AceE[H106M;E401A] generated 50.6 overall volumetric productivity 1.6 pyruvate/L·h yield 0.68 results demonstrate that feasible comparable glucose-based processes.

Amino-acid sequences around two lipoic acid residues in the lipoate acetyltransferase component of the pyruvate dehydrogenase complex of Escherichia coli were investigated. A single amino acid sequence of 13 residues was found. A repeated amino acid sequence in the lipoate acetyltransferase chain might explain this result.
